Induction of increased benzpyrene hydroxylase activity by 2-phenylbenzothiazoles and related compounds.

چکیده
SUMMARY A study of the relationships between structure of 2-phenylbenzothiazoles and related compounds and their ca pacity to induce increased benzpyrene hydroxylase activity in the liver and lung of the rat has been carried out. Thirty-three compounds have been studied. It has been found that intro duction of an appropriate halogen into the 4'-phenyl position approximately doubles inducing activity compared to unsub stituted 2-phenylbenzothiazole. Other substitutions and modi fications of the molecule result in either lesser increases in inducing activity or, in some instances, reduction or total loss of inducing activity. Most compounds show similar inducing effects on both lung and liver. Some, however, do not. Of the more potent inducers, the compound showing this selective effect to the greatest degree is 2-(4'-cyanophenyl)-benzo thiazole, which has more than double the inducing effect on lung as on liver.

Induction of increased benzpyrene hydroxylase activity by flavones and related compounds.
A study of relationships between the structure of flavones, flavanones, and chalcones and their capacity to induce increased 3,4-benzpyrene (BP) hydroxylase activity in the liver and lung of the rat has been carried out. Appropriate halogenation in creases inducing activity. In the compounds studied, hydroxylation reduces inducing capacity, but the corresponding methoxy compounds are active. Tw...
متن کاملInhibition of induction of benzpyrene hydroxylase by nickel carbonyl.
Exposure of rats to nickel carbonyl by inhalation (0.2 mg Ni/liter of air/15 min) or intravenous injection (2 mg Ni/100 gm) was found to inhibit the induction of benzpyrene hydroxylase in lung and liver. Phenothiazine induction and fluorometric measurements of benzpyrene hydroxylase were performed by the methods of Wattenberg and co-workers (31, 32). Benzpyrene hydroxylase activities in lungs a...
متن کاملBenzpyrene hydroxylase activity and its induction by methylcholanthrene in Morris hepatomas, in host livers, in adult livers, and in rat liver during development.
There was no consistency of the induced levels among hepatoma lines, although all the rats were sacrificed exactly 6 hr after onset of feeding under controlled dietary regimens and 24 hr after a single intraperitoneal treatment with methylcholanthrene. Benzpyrene hydroxylase in fetal rat liver was very low in activity, and after birth a marked increase was observed. After administration of meth...
متن کاملSubstrate-induced synthesis and other properties of benzpyrene hydroxylase in rat liver.
The intraperitoneal injection of weanling rats with 1 mg. or less of 3-methylcholanthrene or of certain other polycyclic aromatic hydrocarbons rapidly induces several-fold increases in the activities of two hepatic microsomal enzyme systems which can metabolize 4-dimethylaminoazobenzene and related dyes (I). These TPNH’-dependent systems catalyze the oxidative removal of the N-methyl groups and...
متن کاملBenzpyrene Hydroxylase Activity in Isolated Parenchymal and Nonparenchymal Cells of Rat Liver
Previous studies have implicated the reticuloendothelial cells of the liver in certain aspects of steroid metabolism. The similarity in the metabolism of steroids and polycyclic hydrocarbons suggested that the nonparenchymal cells possibly play a role in these areas. The present study presents evidence that at least one of the microsomal NADPH-requirig enzymes, benzpyrene hydroxylase, is presen...
